I posted a wishlist book this morning.  Member accepted and I was going to mark I will send later.  It did not give me a chance to do that.  It just automatically entered the mail within 2 days.  I won't be able to do that.  Do I mark  it mailed  anyway

Well that's bizarre! Maybe there is a glitch....I've never had a book, once requested of me, skip the screen that has the "I can mail within 2 days" AND the "I can mail later" option.  Keep in mind that even if it says you have 2 days, there is actually a grace period beyond that before the order would be cancelled. When can you mail the book? I would mark it mailed at the latest date and PM the member to explain what happened, that the option to mail later wasn't there, and that you will actually be mailing the book "x" number of days after marked mailed. Communication is the key here, and I think as long as it's only a couple of days beyond and you let the member know it should not be a problem.

I would also notify the PBS team, as there should always be the option of when you want to mail the book when you accept the request.

I second Cheryl's advice - but PM them right away, not after you've waited until the last minute to mark it mailed.  That way they won't wait 5 days thinking 'when the heck is this person going to mail this book?'  I've found that people are generally pretty gracious so long as you give them a heads up.
